Answer: 
The mortar spacers create a 1/4" joint. The maximum recommended mortar joint is 5/8" and the minimum is 1/8." for a joint size other than 1/4", you could create your own spacer or free lay the blocks.

Rated 4.0 out of 5.0 by Glass Block Mortar Spacers work really well These little guys will allow a novice to install glass blocks like a pro. They insure uniformity in the mortar joints. My only problem was when I needed to break off the horizontal legs to use them as end of row spacers. The verticals have a weakened spot to allow them to snap off easily, the horizontals don't have that. I ended up cutting them off with diagonal cutters and smoothing them with a bench grinder. A little more work, but they all worked great in the end and the glass block window looks professional. These seem to me to be a little expensive for what they are, but it's the only game in town.
