What size copper or alum. wire is needed to connect a 100 amp sub panel in the basement.

The run is approximately 70'. The last 15' to the main panel is outside along the house. What size conduit do I need for that as well.

The wire "run" holds a factor to its size. If the feeder wire is running through insulation, the 60'C rating is used. If not running through insulation, the 75'C rating is used. tank less hot water heater sub panel

I am installing a 27 kv tank less hot water system. I need to add a sub panel for the required 40 a breakers (3). I have a 125 a breaker in the main box - is this wire the correct choice to run from the breaker in the main panel to the sub panel?

A 27 kv hot water system? I seriously doubt it. I guess you know that 27 kv would be 27,000 Volts.

Does the jacketed 1/0-1/0-1/0-2 gray stranded al service cable for a sub panel need to be in conduit?

The NEC does not prohibit SER cable from being installed in conduit, subject to local restrictions. Installing SER cable in conduit is difficult and not necessary except in areas where additional protection is required.
