If mold or mildew is present, apply rubbing alcohol (even if this is a new installation). Let the area dry before applying or reapplying silicone. Do not use soap and water to clean surfaces to be sealed because silicone will not adhere to surfaces covered with any soap residues or films that may be left behind.
