No, LED's are not bright enough for us so these are Incandescent
No, these are incandescent lights.
My only thought is that if you bought more than 100 lights, and replaced the twinkle bulb to make them flash, the only strip that would twinkle would be the ones within that 100 bulb circuit. So, you’d have to install one twinkler bulb per circuit strand, etc. If this isn’t the case then I’m stumped as well, I’m sorry.
It's also possible you have a light with a broken lead wire. You can pull out each light that doesn't work and check it, or something like this is a good investment for people with a lot of lights: https://www.homedepot.com/p/LightKeeper-Pro-Light-Tester-1203-CD/100686478?MERCH=REC-_-PIPHorizontal3_rr-_-205786326-_-100686478-_-N

I had the same problem with a 300 light strand this year. I found out that one of the bulbs wasn’t pushed in all the way. Hope this helps you out!
You can make them flash by either buying a special flash/twinkle bulb which is a clear bulb with a painted red tip and place it at the beginning of the string or buy a light controller. The bulb option can be a bit bothersome because it will malfunction rather quickly and stop the flash/twinkle altogether. So your best bet is to buy a fairly inexpensive light controller and go that route; I do hope that helps.
