Spring and early Fall are the best times because thats when grub populations are highest. However, you can apply Milky Spore any time the ground is not frozen. If the ground is frozen the spores will not be able to penetrate the soil and will be blown or washed away. As long as the spore can penetrate the soil it will remain dormant near the grass roots until a grub ingests them while feeding on the roots.
