If I use this to darken an unfinished walnut countertop to be used as a desk, can I seal it with polyurethane or similar so that it doesn't dry out and have to be reapplied?

If you are doing this for a desk skip this product and just go with polyurethane. You can easily use a tinted poly and it will also help your desk move papers smoothly.

If I use this on a new walnut butcher block, will it darken the wood as the walnut stain does? I want it to be a dark walnut finish, but food safe as well. What should I use?

The mineral oil will darken considerably. I would "experiment" on the back side of the countertop so you can see the difference.
