Can this saw be used to cut a vinyl downspout? Both to trim for length and to cut a hole in it?
This saw is probably too coarse to do a good job on cutting a vinyl downspout, especially the hole. I would suggest a finer pitch saw for cutting to length, and a hole saw with a piece of wood for backing for cutting the hole (assuming it is round). A fine pitch blade in a jig saw should also do the job.
Would this saw be able to cut out a hole through a 2x4 board with drilled guide holes?

I believe the saw would work well cutting a tree branch as long as the branch is not too big to be cut safely.
Yes, I have used it to cut small branches (less than 3 inch dia.). It's a very sharp saw.
Definitely, i would say up to 1inch to 3 inch branch would be ideal. Mine is so light and sturdy enough, i take it camping.
